采用~3H-TdR掺入实验观察了D,L-硒代蛋氨酸与亚硒酸钠对猪肾近曲小管上皮细胞(LLC-PK1)DNA合成代谢的影响。结果显示,两种硒化合物对DNA的合成均呈双相效应,在硒浓度为0.5~50μmol/L时,能促进DNA的合成,并且D,L-硒代蛋氨酸的作用比亚硒酸钠更为有效;随硒浓度的增高,两种硒化合物均呈现对LLC-PK1细胞DNA合成的抑制作用,亚硒酸钠的毒性作用明显大于D,L-硒代蛋氨酸,亚硒酸钠浓度为400μmol/L时,LLC-PK1细胞DNA合成的抑制率为94.1%,而D,L-硒代蛋氨酸浓度为400μmol/L时,仅呈现对LLC-PK1细胞DNA合成的轻度抑制作用,与文献结果比较,LLC-PK1细胞对硒的毒性作用有较高的耐受性。
Abstract:The effects of selenomethionine(SeMet) and sodium selenite(Na_2SeO3) on DNA synthesis were studied in pig kidney epithelial cell line (LLC-PK1). Results revealed a biphasic effects of both Na_2SeO3 and SeMet on DNA synthesis. At concentrations between 0.5 and 50μmol/L, both of selenium compounds stimulated cell DNA synthesis and SeMet was more effective. At concentrations of 200μmol/L and above they inhibited cell DNA synthesis, and the toxicity of Na_2SeO3 was greater than SeMet.
