目的 探究白芸豆提取物联合左旋肉碱对肥胖小鼠的减脂功效。方法CD-1雄性小鼠按体重随机分为空白对照组(10只)和模型组(60只),空白对照组喂食维持饲料(脂肪供能比为10%),模型组喂食高热量饲料(脂肪供能比为45%)2周后,根据体重筛选出上2/3的40只肥胖小鼠继续喂食高热量饲料6周,将肥胖模型小鼠随机分为4组:肥胖模型组,低、中、高剂量干预组,每组10只。干预组分别给予白芸豆提取物-左旋肉碱54.8、273.8和2737.8 mg/kg,连续4周每日灌胃,空白对照组和肥胖模型组给予等量生理盐水。观察小鼠体重变化,使用小动物体成分分析仪检测小鼠体成分;采用全自动生化仪检测小鼠血清中总胆固醇、甘油三酯、低密度脂蛋白胆固醇和高密度脂蛋白胆固醇水平;通过苏木素-伊红染色观察脂肪细胞形态和肝脏脂肪浸润。结果 与肥胖模型组比较,各剂量干预组小鼠每日摄食量和饮水量、肌肉量和体内水分含量差异均无统计学意义;小鼠体重、肾周脂肪重量、睾周脂肪重量、肝脏湿重、总脂肪含量、脂肪组织细胞横截面面积均显著降低(P<0.05),肝脏脂肪浸润减少;此外,小鼠总脂肪量与取材称湿重所获得的肾周脂肪重量(r=0.929,P<0.001)和睾周脂肪重量(r=0.931,P<0.001)均呈正向强相关;小鼠血清总胆固醇和甘油三酯显著降低(P<0.05)。结论 白芸豆提取物联合左旋肉碱对治疗小鼠肥胖具有功效,且具有辅助降低血脂的功能。
