摘要:

神经酰胺是一类化学性质活跃的脂类小分子物质,可引起细胞的多种生物学功能改变,如细胞凋亡、衰老、分化、自噬等,然而其作用机制尚不清楚。目前大部分研究证实外源化学物刺激可引起细胞内神经酰胺水平升高,神经酰胺可作为第二信使传递凋亡信号,诱发凋亡。胞内高水平的神经酰胺还可直接作用于线粒体,引起线粒体结构改变和功能障碍。本文综述了在外源化学物诱导细胞凋亡中神经酰胺对凋亡信号转导通路下游相关蛋白的影响,讨论了神经酰胺对线粒体膜通透性、氧化磷酸化、氧化应激、能量代谢障碍和线粒体DNA损伤的作用。
